    The objectivization of evaluation of the dynamics of self-defense instruments

    Název: Objektivizace hodnocení dynamiky úderových prostředků sebeobrany Úvod: Údery horní končetinou představují klíčové prostředky pro systém sebeobrany. Doposud nejsou hodnocené jejich mechanické vlastnosti pro širší skupinu úderů bez ochranných prostředků s uplatněním v tréninkové přípravě, v hodnocení rizik zranění, ve forenzní biomechanice a lékařském právu. Cíle: Cílem této práce je sestavit a objektivizovat hodnocení dynamiky širší skupiny úderů horní končetinou pomocí upřesnění velikosti kontaktní plochy a popisu tlumicích vlastností pěny detekční aparatury. Cílem je hodnotit impaktní sílu, pravděpodobnost přesažení prahové hodnoty pro zlomeninu kostní tkáně, impuls, velikost úderových ploch a tlak pro tři techniky úderu, obě pohlaví a dvě úrovně zkušeností. Metody: Práce se skládá ze tří projektů. Dva kvazi experimenty absolvovalo 169 probandů, mužů a žen, úrovně zkušeností začátečník, pokročilý (n = 169, výška 177,8±9,0; hmotnost 81,6±17,8; věk 27,3±8,1) v rámci jednoho období. Probandi byli měřeni na dvou aparaturách (Kistler, Podokam), kde došlo k záznamu časového průběhu impaktní síly úderu a k fotografickému záznamu tří segmentů horní končetiny. Probandi provedli 3 techniky úderu, celkem 15 pohybových akcí a 9 statických simulací.     Reduced real lifetime of PV panels – Economic consequences

    The maintenance and analyzing failures of PV systems and plants are becoming more and more important issues. Our data from the long-term operation of 85 photovoltaic power plants in central Europe show that their actual lifetime is about half that of the originally planned lifetime. After about 10 years, serious failures of 1st tier (bankable) PV panels occur at an increasing rate. This article presents selected typical data and describes the most serious failures. Furthermore, economic calculations of returns on investment are carried out in relation to the price of electricity, which is currently changing at a rapid pace. It shows that the PV panel lifetime reduction from 20 to 30 years, declared at commercial leaflets, to real lifetime about 10–12 years can reduce PV power plant profit substantially, but the investment is still worth it. The reason is that after 10–12 years ser vice/maintenance expenses to replace damaged PV panels and inverters are growing very quickly. The new information could be helpful for owners of PV power plants to get a more realistic estimation of profits

    A conformational sensor based on genetic code expansion reveals an autocatalytic component in EGFR activation.

    Epidermal growth factor receptor (EGFR) activation by growth factors (GFs) relies on dimerization and allosteric activation of its intrinsic kinase activity, resulting in trans-phosphorylation of tyrosines on its C-terminal tail. While structural and biochemical studies identified this EGF-induced allosteric activation, imaging collective EGFR activation in cells and molecular dynamics simulations pointed at additional catalytic EGFR activation mechanisms. To gain more insight into EGFR activation mechanisms in living cells, we develop a Förster resonance energy transfer (FRET)-based conformational EGFR indicator (CONEGI) using genetic code expansion that reports on conformational transitions in the EGFR activation loop. Comparing conformational transitions, self-association and auto-phosphorylation of CONEGI and its Y845F mutant reveals that Y845 phosphorylation induces a catalytically active conformation in EGFR monomers. This conformational transition depends on EGFR kinase activity and auto-phosphorylation on its C-terminal tail, generating a looped causality that leads to autocatalytic amplification of EGFR phosphorylation at low EGF dose

    Total synthesis of Escherichia coli with a recoded genome

    Nature uses 64 codons to encode the synthesis of proteins from the genome, and chooses 1 sense codon—out of up to 6 synonyms—to encode each amino acid. Synonymous codon choice has diverse and important roles, and many synonymous substitutions are detrimental. Here we demonstrate that the number of codons used to encode the canonical amino acids can be reduced, through the genome-wide substitution of target codons by defined synonyms. We create a variant of Escherichia coli with a four-megabase synthetic genome through a high-fidelity convergent total synthesis. Our synthetic genome implements a defined recoding and refactoring scheme—with simple corrections at just seven positions—to replace every known occurrence of two sense codons and a stop codon in the genome. Thus, we recode 18,214 codons to create an organism with a 61-codon genome; this organism uses 59 codons to encode the 20 amino acids, and enables the deletion of a previously essential transfer RNA

    V této práci navrhujeme postupy pro sestavení kognitivních testů pacientů a jejich vyhodnocení na základě obrazové informace. Druhá část této práce se zabývá praktickou implementací a ověřením těchto postupů. Byly prostudovány nejčastěji používané kognitivní testy a na jejich základě byly navrženy modifikovatelné prvky testových formulářů a jejich grafická úprava. Tyto prvky umožňují dostatečnou variabilitu při tvorbě testů a jejich grafická úprava umožňuje zpětné rozpoznání a vyhodnocení. Součástí práce jsou vytvořené algoritmy pro detekci formulářových prvků a jejich vyhodnocení. Úspěšnost navržených postupů a jejich implementace byla ověřena na reálných datech. Implementace byla provedena v jazyce C# a pro podporu funkce využívá softwarové balíčky třetích stran.In this work we develop methods to design and subsequently process cognitive tests based on image data. Second part of this work deals with implementation and verification of the proposed methods. We examined currently used cognitive tests and designed specific elements for suitable representation of the content of these tests. These elements were graphically designed to be convenient for computer recognition and analysis. This work also includes algorithms for the detection and evaluation of these elements. Results of the implementation of developed methods are presented. The application was implemented in C# programming language and uses third party software packages

The aim was to create a list of products, which are already being manufactured or could possibly be manufactured from this sort of material. Information about the tree species which can be used, about prices and also about the processes of manufacturing for selected types of products is included. The market research was carried out mainly via the internet, partially at the village fairs and markets, eventually by the consultations with experts in the field. The best products in terms of utilization, evaluation and practical processing of the material, are the furniture, pencils made of twigs, and partially also the wooden toys

    The Thesis deals with construction of log furniture made of logs in diameter no more than 10 centimetres which are connected to a round joint and a mortise. This furniture is much popular and required not only for its growing approval rating of the log cabins in this country. The work contains drawing documentation of two chairs and two tables inclusive of a technical description, bill of materials, costing, and their practical production which is given evidence of taking photographs. Afterwards, the copious photographic work section is completed by information about practicable producing procedures, instruments and machines.
